چکیده

Jinten hitam ( Nigella sativa ) is one of the medicinal plants in family Ranunculaceae, native to Mediterranean region and several regions Asia. The seed widely cultivated, distributed, used pharmaceuticals, cosmetics, food industries. Due similar morphology its potential adulterant such as Corchorus spp., N. seeds are susceptible adulteration substitution markets. Molecular markers have become most reliable methods for identification authentication plants. objective this study was select random amplified polymorphic DNA (RAPD) primer generating ). Genomic extracted from samples seed, sp. a mixture both samples. Forty-two RAPD primers were study. A total 227 fragments produced 37 primers, out which 65% polymorphic. Primer OPK-4 OPC-12 generated specific , meanwhile, OPB-1, OPL-5, OPM-3, OPD-5, . molecular marker able using selected primer. This research first report on screening spp.).

An Efficient DNA Isolation Method from Nigella sativa L. (Ranunculaceae) Seeds for RAPD and ISSR Analysis

Nigella sativa L. (Ranunculaceae) seeds (black cumin) that contain high oil, terpene, protein, alkaloids are an economically important for medicine and food industry. Lack of an efficient DNA isolation procedure is a limiting factor for any molecular studies of these plant. We have used a genomic DNA isolation protocol for Nigella sativa seeds based on a hexadecyltrimethylammonium bromide (CTAB...

Analysis of Genetic Variability in Rice Varieties (Oryza sativa L) of Kerala using RAPD Markers

The genetic diversity and phylogenetic relationships of eight different varieties of Rice (Oryza sativa) viz., MO 16, Ptb 43, Palakkaddan Matta, Ptb 40, MO 17, MO 6, Ptb 39 and KTR 2 from Kerala state, South India were analyzed by Random amplified polymorphic DNA (RAPD). Out of twenty 10-mer RAPD primers screened initially, eleven were chosen and used in a comparative analysis of different vari...

GENETIC DIVERSITY ANALYSIS OF RICE (Oryza sativa L.) LANDRACES THROUGH RAPD MARKERS

The molecular marker is a useful tool for assessing genetic variations and resolving cultivar identities. Information on genetic diversity and relationships among rice landraces from Bangladesh is currently very limited. Thirty-five rice genotypes including 33 landraces and 01 HYV of Bangladesh and 1 Indian landrace of particular interest to breeding programs were evaluated by means of random a...

Development of RAPD-SCAR markers for different Ganoderma species authentication by improved RAPD amplification and molecular cloning.

The sequence-characterized amplified region (SCAR) is a valuable molecular technique for the genetic identification of any species. This method is mainly derived from the molecular cloning of the amplified DNA fragments achieved from the random amplified polymorphic DNA (RAPD). In this study, we collected DNA from 10 species of Ganoderma mushroom and amplified the DNA using an improved RAPD tec...

Genetic Variability Evaluation Among Iraqi Rice (Oryza sativa L) Varieties using RAPD Markers and Protein Profiling

Genetic variability and relationships among ten rice varieties were studied by using 20 decamer random primers and SDSPAGE methods. Out of 22, 20 Random Amplified Polymorphic DNA (RAPD) primers revealed polymorphism, while the remaining 2 primers showed no reaction. The primers produced a total of 109 bands, of which 62 (56.88%) were polymorphic. The number of polymorphic fragments for each pri...
